Moxifloxacin can inhibit bacterial DNA synthesis and kill bacteria rapidly. Moxifloxacin can act on bacterial DNA gyrase and topoisomerase IV, resulting in the breaking of enzyme DNA complex. DNA gyrase, also known as topoisomerase II, is composed of gyrA and gyrB subunits. DNA gyrase and topoisomerase I jointly regulate the process of DNA replication. DNA gyrase plays an important role in maintaining the DNA coil in the whole replication process. Topoisomerase IV is composed of Parc and pare subunits. Its structure is similar to that of DNA gyrase. Parc has some homology with gyrA, pare and gyrB. Topoisomerase Ⅳ can distribute the DNA of the progeny to the progeny cells and complete the replication of bacterial DNA together with DNA gyrase. Moxifloxacin can stabilize the enzyme DNA complex in the state after DNA strand cutting and stop DNA replication, thus producing cytotoxic effect. The main action site of moxifloxacin on most Gram-negative bacteria was DNA gyrase, while the main action site against Gram-positive bacteria was topoisomerase IV.
